Ensemble Discovery is developing a new class of therapeutics - the Ensemblins™, orally bioavailable small molecule macrocyclic compounds that have the potential to behave like biologics and can disrupt protein-protein interactions. We have two complementary platforms that provide access to these molecules. Firstly, we use our proprietary DNA-Programmed Chemistry™ (DPC™) platform to generate hundreds of thousands of these molecules for rapid screening. Secondly, we have highly developed methods for the synthesis of Ensemblins as discrete molecules using conventional chemistry. Interrupting protein-protein interactions can provide novel therapies for cancer, inflammation and cardiovascular disease. Our most advanced proprietary programs comprise candidate drugs for inflammation and cancer, targeting particular protein-protein interactions and are at the stage of evaluation in animal disease models. We are also engaged in an ongoing program to screen our Ensemblin libraries against a wide selection of protein-protein interaction targets that will provide small molecule hits against those targets for the first time and represent future opportunities for proprietary or partnered programs. Ensemble was founded in 2004 by Flagship Ventures and David Liu, the inventor of DPC, a Professor of Chemistry and Chemical Biology at Harvard University, and a Howard Hughes Medical Institute Investigator. DPC exerts precise control of chemical reactivity allowing our scientists to program chemical reactions. Ensemble uses DPC to develop Ensemblins ™ as drug candidates and to generate detection signals at the core of our diagnostic assays of protein functional status. Ensemble holds an exclusive, worldwide license from Harvard University to the fundamental intellectual property covering DPC. Since its founding, Ensemble has industrialized the DPC technology and established a dominant intellectual property portfolio covering the technology, including issued patents in the US and Europe. Ensemble has assembled an experienced management team with backgrounds in technology and product development in the pharmaceutical and diagnostic industries plus a strong set of investors including Flagship Ventures, ARCH Venture Partners, CMEA Ventures, Harris and Harris Group, and Boston University.
